Active ingredient and dosage form
Pentoxifylline coated tablets:
1 pill contains 100 mg of Pentoxifylline;
60 tab. packaged.
Pentoxifylline 2% injection in 5 ml ampoules;
1 ml of solution contains 20 mg of pentoxifylline;
10 ampoules per pack.
Mechanism of action
- antispasmodic from the group of purines, has a vasodilating effect, improves microcirculation and rheological properties of blood, increases the concentration of oxygen in the tissues. Enhances the action of antihypertensives and hypoglycemic agents.
Indications and usage
Peripheral blood circulation disorders (endarteritis, Raynaud's disease, etc.); cerebrovascular pathologies (atherosclerotic disorders, ischemic and post-infarction conditions, etc.); vascular pathology of the eyes and inner ear; diabetic nephroangiopathy and other diabetic angiopathies; trophic changes associated with damage to the arteries or veins and impaired microcirculation (postthrombophlebitic syndrome, trophic ulcers, gangrene, frostbite).
Dosage and administration
Pentoxifylline administered intravenously, intraarterially, and inside.
In / in the drug is administered in a dose of 0.1 - 0.3 g (1-3 ampoules) in 250-500 ml of sodium chloride solution or in 5% glucose solution for 1.5 - 3 hours 1 time per day
V / a the drug is administered in a dose of 0.1-0.3 g in 20-50 ml of 0.9% sodium chloride solution for 10-30 minutes 1 time per day.
The course of treatment is 10-12 days, then switch to taking the drug in pills.
Inside appoint 0.2 g (2 tablets) to receive 3 times a day after meals. The enteric-pills are swallowed whole with a large amount of water. If necessary, gradually increase the dose to 1.2 g per day. The course of treatment is 1-3 months.
Gastrointestinal: nausea, vomiting, pain and a feeling of heaviness in the epigastrium, diarrhea.
Cardiovascular: possible tachycardia, palpitation, pain behind the sternum, a sense of heat flow to the face. With IV or intraarterial injection, a decrease in blood pressure is possible.
From the side of the central nervous system: possible headache, dizziness.
Allergic reactions: skin rash, itching, urticaria.
From the blood coagulation system: rarely and mainly in the presence of risk factors - bleeding.
Contraindications
Acute myocardial infarction, severe forms of sclerosis of the coronary and cerebral vessels, massive bleeding, retinal hemorrhage, hemorrhage in the brain, simple diabetic and proliferating diabetic retinopathy; pregnancy; individual intolerance.
Pregnancy and breastfeeding
Adequate and well-controlled clinical studies on the safety of pentoxifylline in pregnancy have not been conducted.
Pentoxifylline and its metabolites are excreted in breast milk. If necessary, use during lactation should stop breastfeeding.
Use with caution in patients with coronary artery disease, arterial hypotension.
In case of impaired renal function or severe hepatic impairment, correction of pentoxifylline dosing regimen is required.
During treatment, blood pressure should be monitored.
When used concomitantly with antihypertensive drugs, insulin, oral hypoglycemic drugs, it may be necessary to reduce the dose of pentoxifylline.
With simultaneous use with anticoagulants, blood clotting rates should be carefully monitored.
Drug Interactions
Pentoxifylline may potentiate the action of antihypertensive drugs.
Against the background of parenteral administration of pentoxifylline in high doses, it is possible to enhance the hypoglycemic effect of insulin in patients with diabetes.
With simultaneous use with ketorolac, there may be an increased risk of bleeding and / or an increase in prothrombin time; with meloxicam - increased risk of bleeding; with sympatholytics, ganglioblokatorami and vasodilators - may decrease blood pressure; with Heparin, fibrinolytic drugs - increased anticoagulant action.
Cimetidine significantly increases the concentration of pentoxifylline in the blood plasma, therefore, with the simultaneous use may increase the likelihood of side effects.
