Composition

Each pill contains as active substance 0.05 g and 0.1 g of Sumatriptan in the form of succinate. Excipients: microcrystalline cellulose, milk sugar, potato starch, sodium carboxymethyl starch, Magnesium stearic acid, hydroxypropyl cellulose or hydroxypropyl methylcellulose, polyvinylpyrrolidone, polyethylene glycol 4000, talc, titanium dioxide.

Pharmacodynamics

Interacts with5-hydroxytryptamine Type I receptors located in the smooth muscles of the blood vessels of the brain selectively constrict the vessels in the carotid artery system, inhibiting the activity of the trigeminal nerve. All this helps to reduce the severity of pain in migraine. The clinical effect is noted after 30 minutes.

Pharmacokinetics

When taken orally, sumatriptan is rapidly absorbed. 70% of the maximum serum concentration is reached after 45 minutes. The average absolute bioavailability is 14% due to presystemic metabolism and incomplete absorption. The degree of binding to plasma proteins is small (14–21%), the half-life is 2 hours. The main metabolite (indolucetic analogue of sumatriptan) is derived mainly with urine in the form of free acid and its glucuronide conjugate.

Indications for use

Relief of acute migraine attacks (with and without aura).

Dosage and administration

Inside The pill is swallowed whole with water. Adults: single dose of 0.05 g, in some cases - 0.1 g. Maximum daily dose of 0.3 g. If the symptoms of migraine do not disappear and do not decrease after taking the first dose of the drug, then take the second dose to relieve the same attack it does not follow. If the symptoms have decreased or disappeared, and then resumed, a second dose can be taken within the next 24 hours. The interval between doses is at least 2 hours

Contraindications

Individual intolerance to the drug, ischemic heart disease, angina pectoris; hemiplegic, basilar or ophthalmoplegic forms of migraine, occlusive peripheral arterial disease, uncontrolled arterial hypertension; a history of stroke or transient cerebral circulation; liver failure, pregnancy, breastfeeding, child age, advanced age (over 65 years), simultaneous use with ergotamine and its derivatives.

special instructions

Amigrenin should not be used prophylactically. Before the appointment of sumatriptan to patients with newly diagnosed migraine or atypically flowing, other potentially dangerous neurological diseases should be excluded. It must be borne in mind that patients with migraine, there is a risk of stroke or transient cerebral circulatory disorders. It should not be prescribed to patients with suspected heart disease without prior examination. The first 2-3 doses of the drug should be carried out under the supervision of a physician (a spasm of the coronary arteries is possible). In patients with hypersensitivity to sulfonamides with the introduction of sumatriptan there is an increased risk development allergic reactions (from skin manifestations to anaphylactic shock). If there is no effect on the first dose, the diagnosis should be clarified. Experience with sumatriptan in patients over 65 years of age is limited (there is no significant difference in pharmacokinetics compared with younger patients).Since the use of Amigrenina can cause drowsiness, care should be taken when driving a car and working with machinery.

Overdose

In case of overdose, the patient should be observed for 10 hours, if necessary, symptomatic therapy.

Drug interaction

Simultaneous reception is not allowed: products containing ergot alkaloids; lithium preparations; inhibitors of the reverse neuronal serotonin uptake; MAO inhibitors.
