Composition

Zuclopentixol decanoate

Excipients: triglycerides.

Mechanism of action

Clopixol depot is an antipsychotic (neuroleptic) drug derived from thioxanthene. It is believed that the antipsychotic action of zuclopentixol is due to the blockade of dopamine receptors in the CNS. Thioxanthene derivatives have high affinity for dopamine D₁- and D₂-receptors.

Zuclopentixol causes a fast, transient, dose-dependent sedative effect, prior to the development of antipsychotic action.

Unlike zuclopentixol hydrochloride, zuclopentixol acetate has a longer duration of action - 2-3 days, and zuclopentixol decanoate is a depot form and its effect lasts for 2-4 weeks.

Indications and usage

Maintenance therapy for schizophrenia and paranoid psychosis.

Contraindications

Acute overdose of barbiturates, opioid receptor agonists, acute alcohol intoxication, coma, hypersensitivity to zuclopentyxol.

Dosage and administration

A single dose is 200-750 mg, the frequency of administration is 1-4 weeks and is determined by the clinical situation.

Adverse reactions

From the side of the central nervous system: dizziness, drowsiness (especially when using the drug in high doses and at the beginning of treatment), the development of extrapyramidal disorders (mainly at the beginning of treatment), disturbance of accommodation; rarely, with prolonged therapy - tardive dyskinesia.

Cardiovascular: tachycardia, orthostatic hypotension.

Gastrointestinal: dry mouth, constipation, rarely - minor transient changes in liver samples.

Urogenital: urine retention.

Drug interactions

With simultaneous use with drugs that have a depressant effect on the central nervous system, with ethanol, means for anesthesia increases their inhibitory effect on the central nervous system, potentiates the effect of means for anesthesia.

With simultaneous use with guanethidine, it is possible to reduce or completely suppress the hypotensive action of guanethidine.

The case of the development of ZNS with simultaneous use with klorazepatom.

With simultaneous use may reduce the effects of levodopa and adrenergic agents.

With simultaneous use of lithium carbonate may increase the side effects of zuclopentixol.

When applied simultaneously with Metoclopramide, piperazine increases the risk of extrapyramidal symptoms.
