Mechanism of action

PRONORA - antiparkinsonian drug, stimulator of dopaminergic transmission in the central nervous system, peripheral vasodilator.
Experimental studies have shown that piribedil improves metabolism in brain tissues by stimulating the transmission of electrical impulses in the cerebral cortex, increasing oxygen consumption, increasing pO2 in the tissues of the cerebral cortex, and improving blood supply by increasing the incoming blood flow. This action is due to the stimulation of central dopamine receptors (since dopamine antagonists, for example, pimozide, are able to inhibit the action of Pronoran).
In humans, piribedil causes dopaminergic stimulation of cortical electrolytic activity, both in wakeful periods and during sleep periods; acts on various functions controlled by dopamine.
By acting on dopamine receptors located in the smooth muscles of the peripheral vessels, it has a vasodilating effect.
It has been shown that in case of intermittent claudiness due to obliterating disease of the arteries of the lower extremities while taking the drug, patients are able to go longer distances.

- as an additional symptomatic therapy for chronic cognitive impairment and neurosensory deficiency in the process of aging (including attention and memory disorders);
- Parkinson's disease in the form of monotherapy (in forms mainly accompanied by tremor) or in combination with levodopa preparations (at the onset of the disease or secondarily in forms accompanied by tremor);
- as an additional therapy for intermittent claudication due to obliterating diseases of lower limb arteries (stage II);
- Ischemic symptoms in ophthalmic diseases.

Dosage and administration

The drug is administered orally at a dose of 50 mg 1 time / day, after the main meal. If necessary, you can increase the dose to 100 mg / day - 50 mg 2 times / day at the end of the main meals.
In Parkinson’s disease, 150-250 mg / day (3-5 tab. / Day) are prescribed as monotherapy, respectively, divided into 3-5 doses. When used in combination with levodopa preparations, a single dose is 100-150 mg / day in 2-3 doses.

Adverse reactions

Gastrointestinal: rarely - nausea, vomiting, flatulence.

From the side of the central nervous system: rarely - anxiety, arousal.

Cardiovascular: extremely rare - orthostatic hypotension.

Special notes

When prescribing the drug to patients with arterial hypertension, it is necessary to simultaneously conduct antihypertensive therapy.
Side effects from the gastrointestinal tract can be reduced or completely eliminated by individual selection of the dose or taking the drug strictly after the main meal.
Digestive disorders occur when an individual predisposition and in the case of taking the drug in between meals.
It should not be prescribed the drug simultaneously with dopamine receptor antagonists.
Influence on ability to drive motor transport and control mechanisms
Pronoran has no effect on the ability to drive a car or other mechanisms.

Overdosage

The use of Pronoran in high doses causes vomiting due to the action of piribedil on the chemoreceptor trigger zone.

The drug should be stored at a temperature not higher than 30 ° C.
