Active ingredient and dosage form
5-NOK coated tablets orange, round, biconvex, shiny.
1 pill contains Nitroxoline 50 mg;
Excipients: lactose, carboxymethylcellulose, sucrose, E110;
in plastic bottles of 50 pcs., in a carton one bottle.
Mechanism of action
5-NOK is an antibacterial drug derived from 8-hydroxyquinoline. Forms complexes with metal-containing enzymes of the microbial cell and blocks its metabolism.
A drug active against pathogens of urinary tract infections - gram-positive, gram-negative bacteria, as well as some fungi.
To drug sensitive Escherichia coli, Ureaplasma urealyticum, Mycoplasma hominis, Candida spp., Torulopsis spp.
To drug impermanently sensitive Proteus spp., Staphylococcus spp.
To drug are resistant Pseudomonas spp., Providencia spp., Klebsiella spp., Enterobacter spp., Serratia spp., Anaerobic bacteria.
The sensitivity of Streptococcus spp., Corynebacterium diphtheriae, Enteroccocus spp., Salmonella spp., Shigella spp., Neisseria gonorrhoeae, Haemophilus influenzae.
Indications and usage
Treatment of acute, chronic and recurrent urinary tract infections caused by gram-positive and gram-negative bacteria and fungi sensitive to nitroxoline:
- pyelonephritis;
- cystitis;
- urethritis;
- epididymitis;
- an infected adenoma or carcinoma of the prostate gland.
Prevention of infections after surgical and diagnostic interventions on the urinary tract (catheterization, cystoscopy,after kidney and urinary tract surgery).
Prevention of recurrent urinary tract infections.
Contraindications
- cataract;
- renal failure severe (CC less than 20 ml / min);
- severe liver failure;
- pregnancy;
- lactation period (breastfeeding);
- hypersensitivity to nitroxoline or other components of the drug;
- hypersensitivity to quinolines.
Dosage and administration
For adults 5-Nok appoint 400-800 mg / day, divided into 4 doses.
The average daily dose is 400 mg. If necessary, therapy continues continuously or courses (2 weeks for a month).
AT severe cases dose can be doubled; however, it should not exceed 200 mg 4 times / day.
Children over 5 years old the drug is prescribed for 200-400 mg / day, divided into 4 doses.
Children weighing more than 8 kg prescribe 200 mg / day, divided into 4 doses.
For patients with moderate renal failure (CC more than 20 ml / min), liver failure the usual dose should be halved (50 mg 4 times / day).
Tablets should be taken before meals.
Adverse reactions
Gastrointestinal: nausea, vomiting (can be prevented by taking the drug during meals); rarely, changes in the level of transaminases and uric acid in the serum.
Nervous system: ataxia, headache, paresthesia, polyneuropathy.
Cardiovascular: tachycardia.
Hemic and lymphatic: thrombocytopenia.
Allergic reactions: rarely, skin rash, quickly passing after cessation of therapy.
Other: Due to the fact that nitroxoline is excreted by the kidneys, urine is stained in an intense yellow-red color.
Pregnancy and breastfeeding
The drug is contraindicated for use during pregnancy and lactation (breastfeeding).
Special notes
Nitroxoline should not be prescribed simultaneously with drugs containing hydroxyquinolines or their derivatives.
Care must be taken when using the drug in patients with impaired renal function due to the possible cumulation of nitroxoline.
With prolonged or repeated treatment with high doses of halogen derivatives of hydroxyquinoline, cases of development of peripheral neuritis and damage to the optic nerve are described. When using nitroxoline such cases were not observed.
You should not assign treatment for more than 4 weeks without additional examination of the liver and kidneys.
